Favism: Impairment of Proteolytic Systems In Red Blood Cells
نویسنده
چکیده
Red blood cells (RBC) from favic patients are characterized by (a) severe oxidative damage (contributed by autoxidation of divicine and isouramil, two pyrimidine aglycones present in fava beans) and (b) greatly increased calcium levels. In vitro, both autoxidation of divicine and calcium loading produced marked alterations of proteolytic systems in intact RBC. Specifically, autoxidizing divicine inactivated procalpain, the proenzyme species of calciumactivated cytosolic neutral proteinase, or calpain. Inactivation was much greater with glucose-6-phosphate dehydrogenase (G6PD)-deficient RBC than with normal RBC. On the other hand, loading of normal and G6PD-deficient RBC with calcium resulted in (a) conversion of procalpain to calpain and eventual autoproteolytic inactivation of calpain itself, and (b) extensive release of acid endopeptidase activity from the membranes into the cytosol. Damaged RBC from favic patients had significantly lowered procal-

متن کاملThe Assessment of Erythrocyte Reduced Glutathione Concentration in the Neonates suffered from G6PD Deficiency
Abstract Background and Objective: Glutathione as a master antioxidant plays a key role in eliminating free radicals from your body and in preserving cell membrane integrity esp. blood cells. We aimed to assess the Erythrocyte Glutathione level of favism and non-favism neonates. Material and Methods: This study was conducted in Ekbatan hospitals of Hamadan (2009) on 15 favism patients and 60...
متن کاملFavism: divicine hemotoxicity in the rat.
Favism is an acute hemolytic anemia known to occur in susceptible individuals who ingest fava beans. Susceptibility to favism is conferred by a genetic deficiency in erythrocytic glucose-6-phosphate dehydrogenase (G6PD) activity. Although the fava bean pyrimidine aglycones, divicine and isouramil, have been implicated in the onset of favism in humans, the lack of a well-defined experimental ani...
متن کاملEvaluation of liver and kidney function in favism patients
Backgrounds: G6PD deficiency is the most common enzymopathy of red blood cells. The clinical symptoms of favism are jaundice, hematuria and haemolytic anaemia that seem to affect liver and kidney in long term. Thus we evaluate kidney and liver function of favism patients in an endemic area of the disease with a high rate of fava beans cultivation. Methods: This study was performed on favism pa...
